Transcriptome profiling combined with network analysis deepens the understanding of immune response mechanisms in blood of pacific oyster Crassostrea gigas infected by Vibrio alginolyticus

Mass mortalities of oysters during the summer have been reported for years. Among biological factors causing oyster death in summer, Vibrio infection is considered to be most important factor. alginolyticus one main pathogens diseases. V. can cause disease and fish, shrimp, shellfish, other aquatic products. Human with also skin infections, external ear canal Hemocytes blood Crassostrea gigas resemble vertebrate macrophages play an role immune regulation organisms. Therefore, it great significance analyze transcriptome study anti-pathogen mechanism C. . In our study, we performed a profile gene expression on infected at 12 48 hours, identified 2,494 1,165 differentially expressed genes (DEGs) two sampling time points. Then, functions DEGs were using GO (Gene Ontology) KEGG (Kyoto Encyclopedia Genes Genomes) functional enrichment analysis, revealing that there multiple significantly enriched terms signaling pathways related immunity. And protein-protein interaction (PPI) network was construct significant immune-related pathways. At last, screened validated key protein numbers or involved more pathways, verified their changes by quantitative RT-PCR (qRT-PCR). For first time, studied defense based network, which explains how lacking specific immunity survive pathogen-rich environment.
